Mouse anti-Human Interleukin-31 (IL-31)-ELISA Set

DESCRIPTION

Intended Use
This set is specifically designed for quantitative determination of human Interleukin-31 (IL-31)
concentrations in cell culture supernatant, plasma and serum.

Background
Interleukin-31 (IL-31) is a 24kD α-helical protein and a member of the IL-6 family of cytokines that is
preferentially expressed by CD4+ T cells[1]. This cytokine elicits its response by binding to a
heterodimeric complex consisting of a gp130-like receptor chain and the oncostatin M receptor
subunit. On target cells which include a broad range of immune and non-immune cells, IL-31 binding
can induce activation of the Jak/STAT, AKT and MAPK signaling pathways. Due to the ubiquitous
expression of its receptor complex, IL-31 has numerous physiological roles that include regulating
hematopoiesis and immune response[2,3]. Over-expression has been linked chronic inflammatory states
such as dermatitis, pruritus, airway hypersensitivity and inflammatory bowel disease[4-6].

Principle of Assay
This assay incorporates a quantitative sandwich enzyme immunoassay technique. Unlabeled
monoclonal antibody specific for human IL-31 is first coated onto 96 well microplate(s). Standards and
samples are then added to the wells and any IL-31 present is captured by the immobilized antibody.
After a wash to remove unbound material, biotin-labeled anti-human IL-31 detection antibody is added
to the wells, followed by a horseradish peroxidase-labeled anti-biotin antibody incubation. After
another wash, TMB substrate solution is added that will result in a blue color proportional to the
amount of bound IL-31. Color development is then quenched and intensity is measured at 450nm.
