Custom Monoclonal Antibody Production Process
Each phase is optional and can be combined in any number of ways.
Antigen Design and Preparation
We accept various antigen types including peptides, recombinant proteins, and purified native proteins. These may be supplied in a variety of different formats such as in solution in a neutral buffer, a soluble lyophilisate, and contained in gel slices or on nitrocellulose blots.

Immunization and ELISA Titration
Animals are immunized and boosted over several weeks. We select the best-responding animal using a titration ELISA and send you the results. Alternatively, you may opt to have the sera shipped to you to conduct the evaluation in your own lab.
Fusion and Screening for Positive Supernatants
The best-responding animal(s) will receive additional boosts, after which we harvest splenocytes for fusion with a selected myeloma cell line. We then screen for positive supernatants using ELISA and provide you with the results. We can also ship you the supernatant from the best hybridoma-containing wells for evaluation in your lab.
Subcloning, Expansion, and Cryopreservation
We obtain monoclonal cell lines by performing limiting dilution of the hybridoma(s). This also helps ensure the long-term stability of the cells. Once monoclonality is achieved, we confirm the isotype and screen for positive clones using ELISA. We send you the ELISA results and/or we can ship the supernatant from the most promising wells to your lab for additional evaluation. The selected clones are then expanded and cyropreserved. We can also provide cell storage services.
We can produce selected clones in vitro (cell culture flask or bioreactor supernatant) or in vivo (mouse ascites with an immunocompromised option). Small or large scale production is available.
Supernatant or ascites may be shipped separately, pooled following the final collections, or pooled and purified utilizing a selection of the following methods:
- Protein G purification
- Protein A purification
- Ammonium sulfate precipitation
- Caprylic acid precipitation
- PEG precipitation
- Affinity purification
- Size exclusion / Gel filtration chromatography
- Endotoxin removal
