Custom Polyclonal Antibody Production Process
Each phase is optional and clients may select only the generation of antisera / ascites or may choose to perform purification and more.
Antigen Design and Preparation
We accept various antigen types including peptides, recombinant proteins, purified native proteins, and cell lysates. These may be supplied in a variety of different formats such as in solution in a neutral buffer, a soluble lyophilisate, and contained in gel slices or on nitrocellulose blots.

Polyclonal Antibody Generation
Antisera and ascites production is available on a large or small scale basis utilizing our standard protocols or a client supplied immunization procedure and schedule. Small pre-immunization serum samples are typically provided for screening. Upon completion of the protocol, the project may be terminated following your approval or continued on a monthly basis.
Antibody Purification
Antisera and ascites may be shipped separately, pooled following the final collections, or pooled and purified utilizing a selection of the methods below:
- Affinity purification
- Protein G purification
- Protein A purification
- Cross-adsorption
- Ammonium sulfate precipitation
- Caprylic acid precipitation
- Size exclusion / Gel filtration chromatography

Antibody Conjugation
SouthernBiotech offers a variety of conjugation options, to support your different assay needs, such as horseradish peroxidase (HRP), alkaline phosphatase (AP), biotin, β-galactosidase and fluorochromes including Alexa Fluor® dyes, FITC, PE, and PE/CY7.
Antibody Testing and Characterization
We can characterize your antibody using any of the following:
- Serum titer evaluation
- ELISA testing for specific activity
- Endotoxin testing by LAL assay
- HPLC
- Western blot
- Flow cytometry
- Immunohistochemistry

| Note - We recommend that multiple animals are used in order to maximize the potential for a robust immune response | |
